Bacterial infectious diseases are a kind of diseases that seriously harm the health of pet poultry. At present, antibiotics are the main drugs used in the clinical treatment of bacterial infectious diseases. However, the abuse of antibiotics leads to the rapid increase of drug-resistant bacteria, especially multi-drug-resistant bacteria, which cannot effectively control infection, becoming a difficult problem in clinical treatment and bringing heavy economic burden to the society.
Bacterial vaccine can improve the resistance of susceptible animals to pathogenic bacteria, reduce the incidence of pathogenic bacteria infection, and is conducive to the control of infectious diseases. Therefore, the development of related bacterial vaccines has been a research hotspot in this field.
Generally, the production process of veterinary bacterial vaccine is strain culture, fermentation, purification, configuration and filling, of which purification is divided into concentration, liquid change, collection, and membrane filtration technology can directly complete these three processes at one time.
Tangential flow filtration technology is a widely used membrane filtration technology, and hollow fiber membrane is an important form of separation membrane. In unit volume membrane components, hollow fiber membrane has the largest effective membrane area, high filtration and separation efficiency, easy to clean, simple structure, convenient operation, and no secondary pollution in the production process, so it is widely used.
The following is the application case of Guidling hollow fiber in thallus purification.
Thallus 1
We purified 700mL of thallus 1 with 94 cm² hollow fibers. During the whole experiment, the recovery rate and membrane flux depletion were investigated.
0-73min is the concentration process, 73-121min is the liquid exchange process, 121-149min is the "over" concentration process. During the whole experiment, TMP was basically stable, the liquid inlet speed was unchanged, and the average material flux was 47LMH.
The 700mL liquid was concentrated and changed to 17mL, about 41 times the concentration, and the bacterial concentration was determined to be 2.83×1010cfu/mL. The total number of bacteria was 4.81×1011cfu, and the recovery rate was 70.1%. After a top washing, increase 20mL liquid, 1.68×1011cfu bacteria total, then the total recovery is 94.6%, industrial production according to the dead volume of the pipeline calculation, as far as possible to increase the number of top washing, generally 3 times can be completely recovered.
Thallus 2
We purified 50mL of thallus 2 with 94 cm² hollow fibers. During the whole experiment, the recovery rate and membrane flux depletion were investigated.
0-7min is the concentration process, 7-27min is the liquid exchange process, and 27-34min is the "over" concentration process. During the whole experiment, TMP was basically stable, the liquid inlet speed was unchanged, and the average material flux was 64LMH.
The 50mL feed liquid was concentrated and changed to 20mL, and the bacterial concentration was determined to be 2.8×108cfu/mL. The total number of bacteria was 5.6×109cfu, and the recovery rate was 80%. After a top wash, increase 20mL liquid, 1.12×109cfu bacteria total, then the total recovery is 96%, industrial production according to the dead volume of the pipeline calculation, as far as possible to increase the number of top washing, general top wash 3 times can be completely recovered.
Tangential flow filtration of bacteria by microfiltration or ultrafiltration is one of the important applications of membrane separation. Compared with the traditional filter plate Z cake filtration and diatomite filtration, the tangential flow filtration method can achieve high throughput, high yield, suitable for large-scale continuous operation, and easy to carry out aseptic operation.
