How To Use Tangential Flow Membrane Filtration To Clarify Samples

When the sample volume is large (hundreds of ml or even increased), the needle filter can not meet the filtration time requirements, or the cost is high due to frequent replacement of the filter. Tangential flow membrane filtration can be used to clarify the sample.

What is tangential flow filtering?

Tangential flow filtration refers to the filtration method in which the liquid flow direction is perpendicular to the transmission direction. Compared with dead angle filtration, the liquid flow direction in tangential flow filtration is parallel to the membrane, which can continuously wash off the deposits on the membrane surface, so as to reduce blockage. Phenomenon, which ensures the treatment speed and prolongs the service time of the membrane.

Two media are mainly used in tangential flow filtration: hollow fiber and membrane packaging. Hollow fiber has unique open channel and moderate shear force, which is very suitable for processing samples with high solid content, high viscosity or shear sensitivity. The clarification and filtration process can be effectively completed by using hollow fiber in the sample before chromatographic separation.

Hollow fiber is used for clarification and filtration. It is not necessary to remove cells or cell fragments by centrifugation in advance, that is, hollow fiber can be directly used to clarify E. coli lysate without hollow fiber. The fermentation broth secreted and expressed by mammalian cells or yeast can be clarified directly without removing cells.
