1.Description
Guidling 5ml centrifugal devices provide fast ultrafiltration, with the capability for high concentration factors and easy concentrate recovery from dilute and complex sample matrices. The available membrane surface area provide fast sample processing, high sample recovery(such as typically greater than 90% of dilute starting solution) Typical processing time is 15 to 25 minutes. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical dead stop in the filter device prevents spinning to dryness and potential sample loss. The ultrafiltrate is collected in the provided centrifuge tube. The 5ml centrifugal devices can be spun in a swinging-bucket (for optimal performance) or fixed-angle rotor. Guidling 5ml centrifugal devices are supplied non-sterile and are for single use only

Guidling centrifugal filter device is supplied with a cap, a filter device, and a centrifuge tube.
Required Equipment
1.Centrifuge with swinging-bucket (preferred) or fixed-angle rotor with wells/carriers that can accommodate 18 mm × 125mm 15 mL conical-bottomed tubes
CAUTION: To avoid damage to the device during centrifugation, check clearance before spinning.
2.Pipettor with 150 microliter (μL) tip for concentrate recovery
3.Choosing the Correct MWCO
Once sample volume is determined, the next step is to select the appropriate MWCO (for ultrafiltration) or pore size (for microfiltration). MWCOs are nominal ratings based on the ability to retain > 90% of a solute of a known molecular weight (in Kilodaltons). Table provides retention characteristics of different MWCO membranes for some solutes. For proteins, it is recommended that an MWCO be selected that is three to six times smaller than the molecular weight of the solute being retained. If flow rate is a consideration, choose a membrane with an MWCO at the lower end of this range (3X); if the main concern is retention, choose a tighter membrane (6X). It is important to recognize that retention of a molecule by an ultrafiltration membrane is determined by a variety of factors, among which its molecular weight serves only as a general indicator.
Therefore, choosing the appropriate MWCO for a specific application requires the consideration of a number of factors including molecular shape, electrical charge, sample concentration, sample composition, and operating conditions. Because different manufacturers use different molecules to define the MWCO of their membranes, it is important to perform pilot experiments to verify membrane performance in a particular application.
Common Variables that Increase Molecule Passage:
1.Linear versus globular molecules.
2.High transmembrane pressure created by g-force in centrifugal concentrators. (This is especially important in the case of linear molecules, for example DNA fragments. Decreasing the g-force can increase retention of molecules by a membrane.)
3.Buffer composition that favors breakup of molecules.
4.pH and ionic conditions that change the molecule
5.Low temperature (5 °C versus 25 °C).
